Abstract To determine the best extraction process of liquiritin from licorice,single factor experiment and orthogonal experiment were designed,and the antioxidant activity of liquiritin was preliminarily studied. Ultrasound-assisted extraction was used as the extraction method,and the yield of liquiritin was used as the evaluation index. The effects of extraction temperature,extraction time,solid-liquid ratio, ultrasonic power,and ethanol concentration on the extraction yield were investigated. On the basis of single factor experiment,the extraction process of liquiritin was optimized by orthogonal design. The antioxidant capacity of liquiritin was determined by DPPH free radical scavenging assay and hydroxyl free radical scavenging assay. The results showed that the optimal extraction condition was obtained at 50% ethanol concentration,solid-liquid ratio 1:10(g/ mL),45 ℃,35 min,and ultrasonic power 350 W. Under these conditions,the extraction yield was 3.46 %. The antioxidant activity study showed that the scavenging rate of liquiritin on DPPH radical and hydroxyl radical was 70.99 % and 20.07 %, respectively. The ultrasound-assisted extraction method can be used for the extraction of liquiritin,and the licorice extract had good antioxidant activity.
