Cloning and analysis of two cytochrome P450 genes from Phenacoccus solenopsis
In order to study the Phenacoccus solenopsis' degradation mechanism to pesticides and other xenobiotics,two Cytochrome P450 genes were cloned by the RACE method.One of them contained 1 536 bp Open reading frame(ORF) that encodes 512 amino acids.Its relative molecular mass was 59 843.5,and the isoelectric point was 8.79.The protein should be stable,with no transmembrane region.It got WXXXR(WKNLR),EXXR(ETLR) and FXXGXXXCXG/A(FGEGPRICIG),a heme binding site,which belongs to cytochrome P450 specific sequence.The ETLR was specific to CYP6 family.Also,the gene got the highest similarity with the insects of the CYP6 A family.The gene was named as PsCYP6a1 by the analysis.Another gene contained an ORF of 1 575 bp fragment,encoding 525 amino acids.The relative molecular mass was 61 246.62,while its theoretical isoelectric point was 8.87.The protein also was stable,with no trans membrane region.As to PsCYP6a1,it also contained the cytochrome P450 conserved sites.So it was named as PsCYP6a2.The nucleotide sequence similarity of the two cytochrome P450 gene was 55.56%,the amino acid sequence similarity of 39.41% residues similar accounted for44.12%.Two P.solenopsis cytochrome P450 genes that belong to CYP6 family were successfully cloned.
Relation between several agronomic traits and pod mass per plant of main peanut cultivars (lines) in Hebei , Shandong and Henan Provinces
In order to provide references for effectively use the right peanut varieties,67 main peanut cultivars and lines were chosen as materials,which were from Hebei,Shandong and Henan province.The relation between the pod yield per plant(PYP) and 8 characters were analyzed seperately,the 8characters included the kernel percent,100-pod mass,100-kernel mass,the days from seedling to 50%flowering(DSF),the days from seedling to maturity(DSM),the main stem height,the No.of fruit branches and the No.of pods.The 67 materials were compared and classified by cluster analysis.The results showed that 2 characters,the kernel percent and 100-kernel mass,were negatively with the PYP,especially the former,which reached a very significant level;the other 6 characters were all positively correlated with the PYP,and their statistically coefficients of correlation reached very significant level but the 100-pod mass and the main stem height.The No.of pods was the most important character in 5 positive characters that improve the PYP,and the kernel percent was the most important character in 3 negative characters which decreas the PYP.The regression equation established by the PYP and the other characters reached a very significant level,it could be used on the forecast of production practice.The third kind of varieties was chosen as high-yield cultivars by cluster analysis.When making breeding objectives,breeders should focus on characters with both high heritability and high correlation with economic characters,and farmers should read the reports of regional trial made by government before choosingthe cultivars.
In vitro rapid propagation of Campanumoea javanica Bl. Stems via multiple shoots using thidiazuron
In order to solve the problem of poor sustainable subculture and repeatability at propagative stage of Campanumoea javanica Bl,the excised stems with buds from sterile seedlings was used as explants,the MS as base medium and supplemented with different concentration of 6-benzyladenine(BA)and thidiazuron(TDZ) to optimize plant growth regulators(PGRs) prescription.The plantlets were obtained by used the methods of root induction directly or indirectly.The results showed that the combination of 0.5 mg/L BA and low concentration of TDZ(0.01-0.05 mg/L) could induce ideal multiple shoots and had the great proliferative rates and probable subculture number.The best PGRs prescription was 0.5 mg/L BA + 0.02 mg/L TDZ,the proliferative rates at the fifth subculture was 10.9 and the probable subculture number reached to 10.The rooting rate and coefficient was 28.5%-30.5% and2.5-3.3 respectively by using the direct rooting method;30%-100% rooting rate and 1-5.3 rooting coefficient was obtained by used the indirect rooting method,when shoots was pre-treated with different concentration of IBA,the rooting rate and coefficient was reached to 100% and 5.3 respectively after the base of excised shoots were dipped in 300 mg/L IBA solution for 15 min.90% of plantlets survived after30 days' acclimatization.The results indicated that the optimal in vitro rapid propagation system of C.javanica Bl via multiple shoots could subculture 10 times abidingly and the propliferative rates could kept10;The indirect rooting method was better than the direct one,and that shoots was dipped in high con-centration of IBA solutions for 15 min at the stage of pretreatment could get the best rooting effect.

Screening, identification and inhibitory effect of antagonistic bacterium on Fusarium oxysporum f. sp. nevium
An antagonistic bacterial strain BTF5 against Fusarium oxysporum f.sp.niveum was isolated from rhizosphere soil collected from continual watermelon mono-cropping by dual-culture,and identified according to morphology,biochemical assay and homology analysis of 16S rDNA sequence.The results showed that inhibition rate of the strain BTF5 against F.oxysporum f.sp.niveum was up to 63.09% in the antagonistic experiment.Compared with sequences listed in the Gen Bank database,strain BTF5 had the highest genetic similarity with Burkholderia vietnamiensis and homology is 99%.Less change in the inhibition effect of antagonizing against growth of F.oxysporum f.sp.niveum was observed during eight generations of transfers on artificial media,indicating its potential biocontrol efficacy and application prospect.
Establishment of shoot regeneration system from leaf explants of strawberry‘Fengxiang’
Using leaf of Fragaria ananassa Duch.‘Fengxiang'(main cultivar variety) of as explant,the survival rate,callus tissue,adventitious bud induction rate and regeneration bud rooting culture treated with different concentrations of growth hormone were studied and the regeneration system was established.The results showed that the medium of MS + 1.5 mg/L Thidiazuron(TDZ) + 0.4 mg/L 3-indolebutyric acid(IBA) was the most suitable for the cultivation,that the regeneration buds had the best rooting effect with medium of 1/2MS + 0.1 mg/L IBA.
Comparison between colloidal gold immunochromatographic strips and HPLC-QTOF-MS for determination of tadalafil in health care products
Colloidal gold immunochromatographic strips and high performance liquid chromatographyquadrupole time of flight-mass spectrometry(HPLC-QTOF-MS) were used to detect tadalafil in health care products and the two methoods was compared.The results showed that the detection limit of colloidal gold immunochromatographic strips for thioaildenafil,acetildenafil,noracetildenafil and pseudovardenafil was 60 μg/mL,and for homosildenafil was 30 μg/mL.The detection limit of HPLC-QTOF-MS for thioaildenafil,acetildenafil,noracetildenafil,pseudovardenafil and homosildenafil was 0.2 μg/mL.The results of two methods determined the actual samples qualitatively were consistent.Colloidal gold immunochromatographic strips had the avantages of ease of use,rapidity and visualization over HPLC-QTOF-MS,could be fit for rapidly qualitative analysis.HPLC-QTOF-MS could be suitable for the validation of positive samples and accurate quantificaton.
Landscape trees counting by remote sensing technology based on UAV
Counting experiments were carried out on Bauhinia variegata planted in the wetland park of South China Agricultural University and Armeniaca mume planted in Luogang Xiangxue Park in Guangzhou city,based on technology combining UAV photography with "crown extraction" in order to introduce UAV and forest remote sensing technology into landscape project to improve work efficiency.Moreover,three steps including aerial photography,image preprocessing and image interpretation were analyzed in order to further improve the accuracy.Results showed that the leakage error of test on B.variegata and A.mume were 25.24% and 11.17%,and the misclassification error were 11.49% and 5.92%;All accuracy was preferable except the leakage error of test on B.variegate was a little higher.All three steps could be affected the accuracy of results.The remote sensing technology based on UAV could be a powerful aid to count trees in landscape projects,and the accuracy could be improved by optimize the workflow.
Analysis on the factors affecting the cultivation of new - type farmers —Based on the data from Guangzhou
428 questionnaires were carried out according to the cultivation of new types of professional farmers in five districts of Guangzhou,and the factors influencing the cultivation efficiency of new professional farmers were analyzed by orderly logistics.Based on quantitative analysis of the characteristics of peasants individual characteristics,family characteristics and cultivating the characteristics of the peasants,it was found that the degree of peasants' education,whether the village cadres,the number of migrant workers,the degree of marketization of production,the applicability of cultivating policies,the cultivation mode flexibility,the effect of cultivating new professional farmers around the city had a significant effect.Therefore,it was necessary to speed up the government in adapting to the development of modern agriculture in the function of change,to strengthen the rural human capital investment precision and feasibility,to improve the follow-up of new professional farmers service and management platform,and to focus on interregional,inter-industry development differences,from the perspective of the cultivation program to adjust,so as to promote the cultivation of new professional farmers to enhance the efficiency.
Influence of age and education level of citrus farmers on the selection of vertical cooperative model
The selection willingness of citrus vertical coordination model was studied in order to find out the problems phenomenon that 'The output of the citrus cultivation is increasing without the income increase' in Guangdong Province and upgrade citrus industry.The citrus farmers' age and education level affected vertical coordination mode was suggested,and analyzed the supposition by correlation and return focus analysis based on questionnaifres.The results showed that when the citrus farmers faced on many kinds of vertical coordination model,the age had a significant negative effect and the education level had a significant positive effect on the vertical coordination model(P0.05).
A new Hedychium cultivar Hedychium'Chengxin'
Hedychium 'Chengxin' is a new Hedychium hybrid which was selected from H.coronarium and H.gardnerianum 'Fen'.The plant of H.'Chengxin' is straight and is 142.2 cm in height with lanceolate leaves and long flowering period.Compared with H.coronarium,the labellum of H.'Chengxin' was ornamented with orange patch and more elegant fragrance.The new cultivar was suitable for open field cultivation in Guangdong area.
Effects of temperature on the CO2 release rate of the 4th instar larvae of Diaphania angustalis
The effects of different temperature on the respiratory metabolism of the 4^th instar larvae of Diaphania angustalis were studied.The insects were collected from Alstonia scholaris L.on the east campus of Guangxi University.The results showed that the CO2 release rate of the 4^th instar larvae of Diaphania angustalis changed obviously in different temperatures.The release rate of CO2 at 15 ℃ and 35 ℃ were significantly different from that at the other temperatures(P〈0.01).In the range of 20 ℃ to 30 ℃,however,the CO2 release rates of the 4^th instar larvae of Diaphania angustalis changed within a small range,which were not significant(P〈0.01).The sensitivity of respiration rate of insects to temperature was also calculated.Within 20 ℃ to 30 ℃,the value of Q(10) was minimum.It was concluded that the temperature range between 20 ℃ to 30 ℃ was the optimal condition for the growth and development of the 4-(th) instar larvae of Diaphania angustalis.
