Purification and identification of Fusarium oxysporum f. sp. cubense race 4 β-1, 6- galactanase

Fusarium oxysporum f. sp. cubense race 4 (FOC4) culture fluid was collected from the liquidmedium containing 1% citrus pectin as a carbon source,and sequentially isolated and purified using PM-10 membrane,ultrafiltration,Sephacry S -100 gel filtration chromatography,and SDS-PAGE electro-phoresis. A protein about 50 kDa was successfully obtained from the crude extract. The peptide mass fin-gerprint of the protein was obtained using mass spectrometry,which matches the data of Fusarium oxyspo-rum beta -1,6 - galactanase (gi: 119507928) phase by comparing the data in the database. The openreading frame of the isolate contains a sequence of 1 263 bp,encoding 420 amino acids. Its DNA se-quence contains 2 introns and 3 exons. The gene was named as Foc4gal1 containing 1 368 bp. The signalpeptide should be the 1 -20 amino acid sequence,the theoretical isoelectric point was 5. 26 and the mo-lecular weight was about 47. 2 kDa. The protein should be a stable protein. Fusarium oxysporum f. sp.cubense race 4 β - 1,6 - galactanase protein was successfully purification and identification. Also,thegene was cloned and sequenced.

Analytical method research of difenoconazole·flusilazole 40% microemulsion by high performance liquid chromatography

A high performance liquid chromatography method for the determination of difenoconazole·flusilazole 40% micro-emulsion was established. The separation and determination of difenoconazole·flusilazole in the samples were conducted by C 18 column with the mixture of methanol and water as mobilephase (85∶ 15,by vol) at the flow rate of 1 mL/min at 220 nm by DAD detector. The results showedthat the retention time of difenoconazole and flusilazole are 6. 320 and 4. 700 min. At the injection con-centration from 10 ~100 mg/L,the linear correlation coefficients for difenoconazole and flusilazole were0. 999 8 and 0. 999 9,the standard deviations were 0. 027 0 and 0. 023 9,the variation coefficients were0. 077 0% and 0. 479 0%,and the average recoveries were 99. 70% and 99. 11%,respectively. In thismethod,difenoconazole·flusilazole with good separation effect,was suitable for quantitative analysis ofactive ingredients in preparation.

Evaluation of mulberry varieties resistance to root-knot nematode disease and screening of control agents

The resistance of 19 mulberry cultivars to mulberry root-knot nematode disease was tested,theresults showed that the cultivar Yuesang C20 was immune to the disease,varieties Yuesang C8,Yuesang87 and Yuesang 44 being highly resistant; Yuesang 85,Yuesang C10,Yuesang C21,Yuesang C19,Yuesang C15,Yuesang C121,Yuesang C43 being moderately resistant; Yuesang 51,Yuesang 119,Yuesang 110,Yuesang C45 and Tang 10 × 109 being moderately sensitive and Yuesang 84,Yuesang120,283 × anti -10 being susceptible. In addition,the control experiment of mulberry root-knot nema-tode disease was carried out in laboratory,the results showed that the efficacy of biocontrol agent Paecilo-myces lilacinus and chlorpyrifos were 75% and 70%,respectively; efficacy of biocontrol bacterium Bacil-lus falciparum was 50%. The results provide theoretical basis for field control work.

Effects of several matrix and NAA on propagation of Ixora coccinea L. by air layering

Ixora plants are an ornamental tropical plant with bright colour,great plant arohitecture andwide range of environmental adaptability. The effect of different substrates and the different concentrationsof NAA on rooting of Ixora coccinea L. were studied by air layering. The results showed that among thefour substrates of peat,perlite,water moss and sawdust,the best rooting effect was water moss with therooting rate of 53. 33%. The average number of roots and average root length in water moss were 3. 03and 3. 54 cm,respectively. In addition,four different concentrations of NAA (50,100,150,200 mg/L)were used to treat the strips. The results showed that 150 mg/L treatment had the best rooting effect.The rooting rate was 86. 67%,the average number of roots was 4. 2,and the average root length was3. 13 cm.

Preparation of citric acid modified starch/PVA film and its application in preservation of mango

A series of citric acid modified starch/polyvinyl alcohol composite membranes were preparedby solution casting method to prepare a new environment ally friendly plastic wrap. Structure and proper-ties of composite membrane was characterized by means of infrared spectroscopy,universal testing ma-chine,moisture permeable machine,light transmittance/haze measuring instrument and other analyticalmethods,and discussed the effect of citric acid dosage on the performance of the composite membranewere discussed. The results showed that the citric acid and starch were esterified and cross-linked. Thecitric acid modified starch could improve the mechanical properties of the composite membrane,reducethe haze,hydrophilicity and water vapor transmission rate of the film,and improve the preservation of thefilm to mango. When the citric acid content was 3. 3%,the mechanical properties were optimized,andthe tensile strength and elongation at break were increased by 42% and 79%,respectively.

Change of water status during olecranon peach storage by LF - NMR technique

The relationship between the total signal amplitude and water content of olecranon peachesduring storage was established by using LF-NRM (low-field nuclear magnetic resonance) technique as y=1. 280 8x -12. 17 (R 2 =0. 982 6). With the passage of time,the moisture content of the three kindsof olecranon peach decreased,with the greatest decrease of the free water,followed by the non-flowingwater and the lowest decrease of the binding water. In addition,the fluidity of the three kinds of waterwas weakened,and the degree of binding water is the greatest. The changes of moisture content and totalmoisture content of the three kinds of olecranon peach were related to respiration intensity and hardness.The increase of respiration intensity was accompanied by the decrease of water content,and the hardnesswas significantly positively correlated with the water content.

Optimization design and test of the pore structure of peanut seed -metering device

In order to solve the problem of poor adaptability of existing peanut seed metering devices toseed size,an a internal filling metering device hole-wheel type peanut seed metering device was de-signed,in which the size of seed nest was controlled by regulator. The single seed rate,replaying rateand missing seeding rate of a single hole were used as test indexes,quadratic orthogonal rotary combina-tion experiment was carried out with the rotational speed of seed metering device,the length of seed nestand the size of seed as experimental factors. The Rresults showed that the order of their effectiveness toSeed-metering Performance was as follows: length of filling hole,seed size,speed of wheel. The parame-ter optimization of the model was that when the rotate speed of seed plate was 40 r/min,the length of fill-ing hole was 9 mm,seed size was 15. 6 mm,the Performance was the best,which leaded to the singleseed rate was 95. 3%,repeat rate was 2. 7% and omitting rate was 2. 2%. The seed meter was suitablefor seed size through adjusting the length of filling hole control the seeding capacity.

Simulation analysis and prototype test of air - suction cleaning device to separate camellia seed and camellia shell

An air-suction cleaning device was designed to solve the problem in separation of camelliaseed kernel and camellia shell after dehulling camellia seed. The flow model of cleaning device was setup by using ANSYS/Fluent as tool,then the motion trajectories of camellia seed kernel,broken kernel,big shell and small shell were simulated. The simulation results showed that the materials have threestates in air flow field such as sucked out,retention and drop down. The time of small shell staying in thecleaning device is random,and path length increases with the rotational speed of blower. When the rota-tional speed of blower is 1 600 r/min,small shell will be separated effectively,some big shells will besucked out,but seed kernel and broken kernel will not be sucked out. The prototype test showed that thecleaning rate increases with rotational speed of blower,when rotational speed is 1 600 r/min,cleaningrate will reach to 98%.

Study on the morbidity of an extended grey prediction model

To solve the morbidity problem of an extended grey prediction model in the process of parame-ter identification,the morbidity degree of the model was calculated by the condition number of matrixspectrum,and the main factors that affect the morbidity of the model were summarized. It was concludedthat there was no serious morbid character in some cases. Finally,the results of an example showed thatif the numerical value of original data were too large,the significant disturbances in the accuracy of themodel that caused by the small error of the data could be avoided by multiplying transformation on originalsequence.

Firstly,a systematic way of constructing mutually unbiased unextendible maximally entangledbases (MUUMEBs) was given in bipartite quantum system C d ×C 3l d'from MUUMEBs in C d ×C d' (d' = kd+ r). Then,the result showed that the pair of MUUMEBs in C 4 ×C 15 could be obtained by constructinga pair of MUMEBs in C 4 ×C 5 . Finally,the method of constructing the MUUMEBs was generalized to C d×C el d'in the bipartite quantum system C d ×C 2l d' .

Effect of penicillin spraying against citrus Huanglongbing bacterium Candidatus Liberibacter asiaticus in citrus plants

Citrus Huanglongbing (HLB),the most devastating citrus disease worldwide,restricts the de-velopment of the citrus industry severely. In order to investigate the control effect of penicillin on CitrusHLB,the test was conducted by means of spraying to treat diseased trees which were infected by bacterialpathogen Candidatus Liberibacter asiaticus (Las). The real-time fluorescence quantitative PCR tech-nique was used to monitor the amount of Huanglongbing pathogen in diseased trees dynamically and deter-mine the relative control effect of penicillin on Las. The results showed that the content of Las in diseasedtrees at the 3rd and 7th months after continuous spraying of penicillin 4 times was up-regulated. At thesame time,the relative control effects of the penicillin solution were - 170. 16% and - 63. 96%,re-spectively. The above results indicate that the penicillin treatment has no effect on HLB.

The application of woody plants in constructed wetland

In the wetland system,woody plants generally have more biomass and longer life than herbs,and have stronger bioaccumulation ability than organic matter,nitrogen and phosphorus. If the woodyplants could be adapted to the water environment could be applied to the constructed wetland,and thelayered plant communities are constructed,the complicated maintenance work in the later stage of theconstructed wetland could be avoided and the cost of wetland management will be reduced. It could alsoovercome the niche vacancy caused by the seasonal blight of the traditional constructed wetland,maintainthe effect of sewage treatment,and ensure the durability of the operation of the constructed wetland. Thisaccords with the construction idea of low maintenance and durable constructed wetland,and it was worthpopularizing in the constructed wetland configuration. The research of woody plants in constructed wet-land is reviewed,and the prospect of application of woody plants in constructed wetland was prospected.

The effective control and treatment of hyperlipidemia ingredients in 11 common food and foodhomologous materials such as kudzu root,dandelion,mulberry leaf,cassia seed,hawthorn,yam,peril-la,lotus leaf,raisin,coix seed,and monacolin was elaborated. The mechanism of action of loweringblood lipids was explored; the research and development status of hypolipidemic foods based on homolo-gous Chinese medicines was reviewed,and the problems that need to be further studied and urgently nee-ded in the development of medicines and foods was pointed out. It provides a theoretical reference for thedevelopment and utilization of medicine and food homologous lipid-lowering foods.
