Abstract Free-hand sectioning is often used in the study of plant cell structure. However, the traditional method of free-hand sectioning needs high technical requirements and complicated destaining and staining. The free-hand sectioning to make slices of rice leaves was improved using two pieces of shaver, and the cell structure was observed through visible and ultraviolet light microscope. The result showed that the clearest cell structure and vascular bundle of leaves were obtained under blue fluorescence, which is fit to analysis the number and size of bulliform cell and cell in vascular bundle. Our results indicated that fluorescence microscopic observation for free-hand section was a very simple and so-easy learned technique.
