Show Abstract ( 691 )
The seedlings germinated from mature seeds of Zingiber mioga(Thunberg) Roscoe were used as explants and the Murashige and Skoog(MS) was employed as base medium supplemented with different concentration of 6-Benzylaminopurine(6-BA), Kinetin(KT), Thidiazuron(TDZ), and 1-naphthylacetic acid(NAA) to study the effects of these plant growth regulators(PGRs) on shoot proliferation times and one-step seedling formation. The result showed that low concentration TDZ(0.01 mg/L) had best effect on shoot multiplication, the shoot proliferation rate was 9.0. The most suitable 6-BA concentration was 2.0 mg/L, the shoot proliferation rate was 7.28. KT had the worst multiplication effect, and the highest proliferation rate was 4.50 on MS+3.0 mg/L KT. However, although MS+0.01 mg/l TDZ are best for shoot multiplication, low rooting rate(38%), mean shoot length(2.28 cm), mean rooting number(0.80) and mean root length(0.52 cm) were obtained in this medium, the regenerated seedlings from the medium failed to meet requirement of acclimatization. MS+2.0 mg/L 6-BA exhibited good growth condition among all growing indexes such as mean shoot length(5.20 cm), rooting rate(100%), mean root number(9.20) and mean root length(4.08 cm). MS+2.0-4.0 mg/L 6-BA and MS+3.0-4.0 mg/L 6-BA+0.01 mg/L NAA were suitable for one-step seedling formation.

Show Abstract ( 438 )
The cuttlefish Sepia pharaonis, an important fishery species in tropical archipelagic environments is highly required in both local and international market trades. Therefore, intensive efforts are currently undertaken to develop practical effects of fundamental physical and chemical parameters on the hatching rate of S. pharaonis fertilized eggs. In this report, the effects of three environmental factors that including temperature, salinity, and pH value were investigated, to understand the suitable ecological conditions for S. pharaonis hatching and enhancement of stocks to provide necessary data for the artificial breeding of S. pharaonis. The present study was conducted following a factorial design to evaluate the appropriate breeding parameters of cuttlefish at different temperatures(18, 21, 24, 27, 30 and 33 ℃), various salinity at(15‰, 18‰, 21‰, 24‰, 27‰, 30‰, 33‰ and 36‰) and at altered pH ranges(7.0, 7.5, 8.0 and 8.5). Results indicate that different temperatures had significant effects(P<0.05) towards hatching of fertilized eggs at 21-30 ℃. With the increase of temperature, the incubation period was significantly shortened, and the optimal temperature was 27 ℃ with the hatching rate of 72.2%. The salinity has a significant effect on the hatching rate of S. pharaonis fertilized eggs at 27‰-36‰, and the optimum salinity ranged from 30‰-33 ‰ with the hatching rate of 70%-80%. By contrast, the effect of pH value on the hatching rate of S. pharaonis was not significant(P>0.05), and the hatching rate was all greater than 70% under the pH values of 7.0-8.5. Regardless of pH, the temperature and salinity are critical factors affecting the hatchability performance of S. pharaonis fertilized eggs. Collectively, the present findings suggest that the artificially hatching conditions for the fertilized egg of S. pharaonis could be set as 24-27 ℃, 30‰-33‰ and pH value at 7.0-8.5 and these patterns may prove helpful in the maintenance of hatchery conditions for aquaculture purposes.

Show Abstract ( 384 )
Magang geese with 0 days old were used to investigate the influences of different light colors on growth performance and some hormones in this study. The white LED light(control group), 480 nm blue LED light and 660 nm red LED light was used in the experiment. In the experimental 7, 35 and 63 days, the body weight and body measurements of geese were recorded, and plasma samples were collected to measure related hormone levels. The results showed that in the late growth stage of geese, the body weight in the blue light was significantly lower than those in red lights(P<0.05). For body measurements, only the breast depth was extremely significantly different throughout the whole experiment period(P<0.01). Extremely significant differences were found in body slope length at day 7, back width at day 35, and shank circumference at day 63 between the three light treatment groups(P<0.01). Adrenalin(ADR) in red light was significantly lower than those in other two groups while growth hormone(GH), insulin-like growth factor 1(IGF-1) and thyroid hormone(T3) had no significant differences among these three groups(P>0.05). The results showed that the sensitivity of geese to blue light and red light was different. In the late growth stage, blue light inhibited the growth performance of geese while red light promoted the growth performance compared with white light.

Show Abstract ( 359 )
Huanglongbing is a devastating disease of citrus, which have caused great loss in the citrus industry of the world. PCR is a reliable method for the diagnosis of Huanglongbing and has advantages such as high sensitivity, strong specificity, high-speed and so on. Previous references have reported the detection PCR primers of Huanglongbing, but there is no report on the comparative analysis of the sensitivity of Huanglongbing specific primers. In order to compare the sensitivity of three pairs of previous reported Huanglongbing specific primers, ten infected Citrus reticulate samples were selected as the PCR reaction template. Specific primers were used to amplify the total DNA of those samples, under the same template and amplification conditions. The results showed that P1/P2 primer has the lowest sensitivity and the DNA samples have not been amplified. The primer P3/P4 has good sensitivity and the amplification band is obvious, but the amplification band of individual samples is not obvious; The primer OI1/OI2 c has the best sensitivity and all sample amplification bands are obvious.

To investigate the optimal reaction conditions for the preparation of methoxy functionalized MQ silicone resin( MMQ),The MMQ was synthesized through the hydrosilylation reaction of Methyl-vinyl containing MQ silicone resin( MVMQ) and trimethoxy( TMOS) siloxane using H2PtCl6 as a catalyst. The conversion of vinyl groups in MVMQ was determined by means of measuring the mass fraction of unreacted vinyl groups using iodometric method. Then, the effects of reaction temperature, reaction time, catalyst dosage, and molar ratio of Si—H/Si—CH=CH2 on conversion ratio of vinyl groups were investigated. By fourier transform infrared spectrum( FT-IR) and nuclear magnetic resonance(~1H-NMR),the structure of the products were characterized. It showed that the MMQ was synthesized successfully and the optimal reaction conditions were reaction time of 6 h,reaction temperature of 80 ℃,catalyst dosage of 10 μg/g( relative to quality of MVMQ),and molar ratio of Si—H/Si—CH=CH2 of 1. 3∶ 1. The conversion rate of vinyl groups in MVMQ was 78. 58%.

Laboratory experiments were conducted to identify the growth of algae response to different spatial size(pilot incubator with volume of 200 L and beaker with volume of 200 mL). algae samples were isolated from a rural reservoir in South china and then inoculated into 4 sets of containers(fertilized beaker, un-fertilized beaker, fertilized tank and un-fertilized tank, respectively)under identical light intensity, temperature and stirring intensity conditions. Chlorophyll-a(chl-a) concentration, algae density were measured and growth curve of algae were protracted during culture period. According to our results: Chl-a concentration was more than 20 000 μg/L and the abundance of cyanobacteria was up to 96% in fertilized beaker, and the chl-a concentration in un-fertilized beaker also reached a high level of 12 000 μg/L. In contrast, there were no evidently gaps among the tanks(either fertilized or un-fertilized) of which maximum Chl-a concentration had only 49 μg/L and 63 μg/L respectively, and the population structure was similar to raw water. It is apparent that the spatial factors was crucial to control algae biomass, and the F value of nutrition was only 1.81 in comparison to 22.32 of that of the spatial factor according to variance analysis. Difference in species diversity, ecological stability, environmental resilience responses to spatial size can mainly explain for the great discrepancy of algae growth level in beaker and tanks. The results of the present study clearly show that spatial scale has an immeasurable influence on the formation of algae blooms.
